de pacientes) y a hormono-resistencia in vitro
resistencia a fulvestrant

PI3K no re-sensibiliza xenoinjertos palbociclib-resistentes a CDKi
La combinación PI3Ki + Fulvestrant es activa en
modelos resistentes a Palbociclib + Fulvestrant

Biological objective: To characterize the molecular patterns of resistance (with a special focus onRb status) upon progression to palbociclib plus endocrine therapy in patients who previouslyachieved clinical benefit with the combination. The biological markers analyzed must cover theentire CDK4/6-Cyclin D-Rb axis, including Cyclin D and E, as the latter appears to play animportant role in the mechanism of resistance to palbociclib
Clinical objective: To define the clinical activity of the combination of palbociclib and endocrinetherapy after prior progression to palbociclib in endocrine-sensitive patients

• Diversos estudios in vitro demuestran que existe
resistencia cruzada entre los diferentes inhib. CDK4/6
• El bloqueo de la vía PI3K/PDK1/mTOR (±CDK4/6i) es una
estrategia válida en modelos resistentes a inhib. CDK4/6
1. Fulvestrant
2. Exemestano + Everolimus
3. Fulvestrant + Everolimus
4. Fulvestrant + Inhibidor α-selectivo PI3K
5. Otra terapia hormonal (Tamoxifeno…)
6. Quimioterapia
